What's wrong:

What's wrong can be used when there's a weird or a not usual situation and you want to know what's happening.

Example:
Your friend is screaming on the phone, you go to him and ask:

Q: What's wrong?

A: Oh, it's nothing. It's just my brother bothering me




What's wrong with you:

You know when you barely know someone and this person comes and asks things about you life?

Example:
Q: Tell me, what do you do for living?

A: What's wrong with you? I don't have to tell you.

Careful:

When you use this "what's wrong with you" it may sound rude depending on how you say it. So if you don't want to sound like that, you can answer:
- "I'm sorry, what do you mean?"
